Collagen metabolism in folic acid deficiency.

چکیده
I . The effect of folic acid deficiency on collagen formation, with respect to the level of collagen-protein synthesis and the extent of collagen-proline hydroxylation, has been studied. A folic acid-free diet containing 10 g sulphasuxidine/kg was used to induce folate deficiency in young male rats. Animals were judged folic acid-deficient on the grounds of retardation of growth, characteristic haematological changes and the urinary excretion of formimino-L-glutamic acid. 2. From isotope incorporation studies it was found that folic acid deficiency caused a marked impairment in collagen synthesis. It was shown, however, from the use of pair-fed control animals, that this was due in part to a reduced food intake accompanying the vitamin deficiency. The further reduction in synthesis in addition to that attributable to inanition was considered to arise from an involvement of folk acid in general protein synthesis, since it was found that the synthesis of elastin and non-collagenous skin proteins was similarly impaired. 3. Reduced synthesis of collagen was not considered attributable to lack of hydroxylation of peptidyl proline that may occur in ascorbic acid deficiency. Hydroxylation of collagenand elastin-proline was only very slightly impaired in folic acid deficiency. It was concluded that, at least when ascorbic acid is present, folk acid is not directly essential for the hydroxylation, in vivo, of peptidyl proline. The results support the view that ascorbic acid participates direct, in vivo, in this hydroxylation rather than indirect by simply maintaining an adequate level of reduced folates. They do not, however, exclude the possibility, arising from the known ability of tetrahydrofolate to participate in the hydroxylation of collagen proline in vitro, that when both ascorbate and folate are present, the latter may, in some measure, share in the role of reducing agent in the hydroxylation of peptidyl proline in vivo.

This study presented a mini review on folic acid deficiency and recommendations for its supplementation in thalassemia intermedia (TI). TI is a clinical condition which lies between thalassemia major and thalassemia minor. Although TI patients may not need regular blood transfusion, precise diagnosis and management are critical for the prevention of clinical complications and quality of life im...
متن کاملComparison of Serum Folic Acid Level in Oral Lichen Planus Patients and Healthy Subjects
Background and aim: Oral lichen planus (OLP) is a chronic mucocutaneous inflammatory disease. Although the etiology of this disease is unknown, some factors including stress, antioxidant deficiency, and folic acid deficiency have been claimed to have a role in its incidence. The purpose of this study was to assess and compare the serum folic acid level in OLP patients and healthy subjects. Mat...
متن کاملInterrelations of vitamin B12 and folic acid metabolism: folic acid clearance studies.
The studies of many investigators have led to a unified concept of the megaloblastic anemias as a single morphologic entity due to defective nucleoprotein synthesis from various causes. The vast majority of patients with megaloblastic anemia have been found to have deficiency of vitamin B12, of folic acid, or of both vitamins. For this reason, the possible interrelations of these two vitamins h...
متن کاملThe urinary excretion of formic acid and formiminoglutamic acid in folic acid deficiency.
In previous investigations it was shown that in folic acid deficiency there is a large increase in the urinary excretion of formimino-n-glutamic acid (l-6). This increase has been explained by recent enzymatic studies demonstrating that the first step in the metabolism of formimino+glutamic acid is the transfer of the formimino group to tetrahydrofolic acid (7-9). Formimino-n-glutamic acid is a...
